Drug intelligence / Profile preview
Cemsidomide plus elranatamab is an investigational combination regimen being developed for the treatment of relapsed/refractory multiple myeloma (RRMM). Cemsidomide (CFT7455) is an orally bioavailable small-molecule molecular glue degrader (MonoDAC degrader) that targets the transcription factors IKZF1 (Ikaros) and IKZF3 (Aiolos) via cereblon (CRBN) E3 ligase modulation, leading to direct anti-myeloma activity and T-cell activation. Elranatamab (ELREXFIO) is an FDA-approved bispecific T-cell engager (TCE) targeting B-cell maturation antigen (BCMA) on myeloma cells and CD3 on T cells, redirecting T-cell-mediated cytotoxicity against the tumor. The combination of cemsidomide and elranatamab, often administered alongside dexamethasone, is designed to synergistically enhance anti-myeloma efficacy by combining direct tumor cell degradation and robust T-cell activation with targeted T-cell redirection. The regimen is currently being evaluated in a Phase 1b clinical trial (NCT07280013 / CFT7455-1102) sponsored by C4 Therapeutics, with Pfizer supplying elranatamab.
