Overview

Centhaquine is a synthetic small molecule cardiovascular drug and first-in-class resuscitative agent approved in India for the treatment of hypovolemic shock. It acts as a vasopressor by selectively activating α2B adrenergic receptors on venous smooth muscle to induce venous constriction and increase venous return to the heart, thereby augmenting cardiac output and improving tissue perfusion. Centhaquine also has activity at central α2A adrenergic receptors, reducing sympathetic drive and decreasing systemic vascular resistance. This dual mechanism enhances tissue blood perfusion while minimizing the risk of arrhythmias due to lack of beta-adrenergic receptor activity. Clinical studies have shown that centhaquine improves blood pressure, base deficit, lactate levels, and reduces mortality in hypovolemic shock patients[1][5][6][7].
