Overview

Cetrelimab + erdafitinib is a combination therapy under investigation for the treatment of advanced or metastatic urothelial carcinoma and other solid tumors. Cetrelimab is a monoclonal antibody that targets the programmed cell death protein 1 (PD-1), acting as an immune checkpoint inhibitor to enhance anti-tumor immune responses. Erdafitinib is an orally administered small molecule that acts as a potent and selective pan-fibroblast growth factor receptor (FGFR) tyrosine kinase inhibitor, targeting FGFR1, FGFR2, FGFR3, and FGFR4. This combination aims to leverage both direct inhibition of tumor growth via FGFR blockade and stimulation of anti-tumor immunity through PD-1 inhibition. The combination has shown manageable safety in early-phase clinical trials and demonstrated improved efficacy over monotherapy in phase 2 studies for metastatic urothelial carcinoma with susceptible FGFR genetic alterations[5][8].
