Cetrorelix + letrozole is a combination therapy investigated for the prevention and management of Ovarian Hyperstimulation Syndrome (OHSS), a potentially life-threatening complication of controlled ovarian stimulation in assisted reproductive technology (ART). Cetrorelix is a synthetic decapeptide that acts as a gonadotropin-releasing hormone (GnRH) antagonist, providing rapid suppression of luteinizing hormone (LH) and follicle-stimulating hormone (FSH) secretion. Letrozole is a third-generation, non-steroidal aromatase inhibitor that blocks the conversion of androgens to estrogens. The rationale for this combination is to synergistically lower serum estradiol levels and downregulate vascular endothelial growth factor (VEGF) expression, thereby reducing vascular permeability and fluid shifts associated with OHSS. This specific regimen has been primarily developed and studied by researchers at the Wuhan University Renmin Hospital (Hubei General Hospital).
