Overview

Cetuximab + bevacizumab + gemcitabine is a combination regimen consisting of **cetuximab** (a monoclonal antibody targeting epidermal growth factor receptor [EGFR]), **bevacizumab** (a monoclonal antibody targeting vascular endothelial growth factor [VEGF]), and **gemcitabine** (a nucleoside analog chemotherapeutic agent). Cetuximab blocks EGFR-mediated signaling and proliferation; bevacizumab inhibits angiogenesis by blocking VEGF; gemcitabine acts as a cytotoxic agent by inhibiting DNA synthesis. This combination has been investigated in clinical trials for treatment of advanced and resected pancreatic carcinoma, as well as metastatic colorectal cancer, with studies indicating that dual EGFR/VEGF pathway inhibition, even when combined with gemcitabine, did not produce substantial efficacy over existing standards, but demonstrated manageable toxicity profiles[1][5][7].
