Cetuximab + cilengitide is an investigational combination therapy consisting of two agents: - Cetuximab, a chimeric monoclonal antibody targeting the epidermal growth factor receptor (EGFR), inhibits EGFR-mediated signaling pathways involved in tumor cell proliferation and survival. - Cilengitide, a cyclic RGD pentapeptide, is an integrin inhibitor that selectively blocks αvβ3 and αvβ5 integrins, disrupting tumor angiogenesis and cell adhesion. This combination has been studied primarily in conjunction with platinum-based chemotherapy (such as cisplatin and 5-fluorouracil) for the treatment of recurrent or metastatic squamous cell carcinoma of the head and neck (SCCHN) as well as advanced non-small cell lung cancer (NSCLC). The rationale for combining these agents is to simultaneously inhibit EGFR-driven tumor growth and integrin-mediated processes critical for tumor progression[1][2][4].
