Cetuximab + cixutumumab is a combination of two monoclonal antibodies used in clinical trials for cancer therapy. Cetuximab targets the epidermal growth factor receptor (EGFR), blocking ligand binding and inhibiting downstream signaling pathways involved in cell proliferation, survival, angiogenesis, and metastasis. Cixutumumab targets the insulin-like growth factor-1 receptor (IGF-1R), preventing its activation by endogenous ligands and thereby inhibiting IGF-1R-mediated tumor cell growth and survival. This combination has been investigated primarily in recurrent/metastatic head and neck squamous cell carcinoma (HNSCC) and advanced non-small cell lung cancer (NSCLC). Clinical studies have not demonstrated significant efficacy or improved outcomes compared to standard therapies; some trials were terminated early due to lack of benefit or safety concerns[1][2][7].
