Overview

Cetuximab + docetaxel + cisplatin + fluorouracil is a multi-agent chemotherapy regimen used primarily for the treatment of locally advanced or recurrent/metastatic squamous cell carcinoma of the head and neck. This combination builds on the standard TPF regimen (docetaxel, cisplatin, and 5-fluorouracil) by adding cetuximab, an epidermal growth factor receptor (EGFR) inhibitor. Cetuximab is a monoclonal antibody that blocks EGFR-mediated signaling pathways involved in tumor growth and survival. Docetaxel is a taxane that inhibits microtubule depolymerization, leading to cell cycle arrest; cisplatin forms DNA crosslinks causing apoptosis; and 5-fluorouracil inhibits thymidylate synthase, disrupting DNA synthesis. The addition of cetuximab aims to enhance antitumor efficacy over standard chemotherapy regimens[1][2][5].
