Overview

Cetuximab + fruquintinib + sintilimab is an experimental combination therapy being investigated for the first-line treatment of patients with pMMR, RAS/BRAF wild-type unresectable metastatic colorectal cancer. This regimen consists of cetuximab beta (an anti-EGFR monoclonal antibody), fruquintinib (a selective small-molecule inhibitor of VEGFR-1, -2, and -3), and sintilimab (a monoclonal antibody targeting the PD-1 receptor). By combining these three agents, the therapy aims to simultaneously inhibit epidermal growth factor signaling, block tumor-associated angiogenesis, and restore T-cell mediated anti-tumor immunity. The combination is currently being evaluated in a Phase 2 clinical trial sponsored by Zhejiang University to determine if this chemotherapy-free approach can provide clinical benefit in the first-line setting.
