Cetuximab + magrolimab is an investigational combination therapy consisting of two monoclonal antibodies with distinct mechanisms of action. Cetuximab is an epidermal growth factor receptor (EGFR) inhibitor used primarily in the treatment of KRAS wild-type colorectal cancer and head and neck squamous cell carcinoma. Magrolimab is a monoclonal antibody targeting CD47, a "don't eat me" signal that allows tumor cells to evade macrophage-mediated phagocytosis. By combining these agents, the regimen aims to both inhibit tumor cell proliferation via EGFR blockade and enhance immune-mediated clearance through CD47 inhibition. This combination has been studied in phase 1b/2 clinical trials for advanced colorectal cancer (including both KRAS mutant and wild-type) as well as in recurrent/metastatic head and neck squamous cell carcinoma[1][3][4][5]. The combination has shown tolerability with potential antitumor activity in heavily pretreated patients.
