This is a combination regimen consisting of two anti-EGFR monoclonal antibodies—cetuximab and panitumumab—administered together with the chemotherapy protocol known as mFOLFOX6. Cetuximab and panitumumab both target the epidermal growth factor receptor (EGFR), inhibiting downstream signaling pathways involved in cell proliferation and survival. The mFOLFOX6 regimen includes folinic acid (leucovorin), fluorouracil (5-FU), and oxaliplatin, which act as cytotoxic agents to disrupt DNA synthesis and repair in rapidly dividing cancer cells. This combination has been studied primarily in metastatic colorectal cancer, particularly in patients with wild-type KRAS tumors[1][2][7]. The use of both anti-EGFR antibodies together is not standard clinical practice due to overlapping mechanisms, increased toxicity risk, and lack of evidence for added benefit over single-agent anti-EGFR therapy combined with chemotherapy.
