Overview

Cetuximab + nivolumab is a combination of two monoclonal antibody therapies: cetuximab, an epidermal growth factor receptor (EGFR) inhibitor, and nivolumab, an immune checkpoint inhibitor targeting programmed death-1 (PD-1). This combination has been studied primarily in recurrent or metastatic head and neck squamous cell carcinoma (HNSCC), where concurrent administration is hypothesized to provide synergistic anti-tumor effects by activating both innate and T-cell dependent adaptive immune responses. Cetuximab inhibits epidermal growth factor-mediated signaling that drives tumor growth, while nivolumab restores anti-tumor immune surveillance by blocking immune inhibitory PD-1 receptor signaling[1][5]. The combination has been evaluated in clinical trials, especially as a second-line or beyond therapy in patients with HNSCC, and in experimental settings for other advanced solid tumors[1][6][8]. Both agents are typically administered intravenously.
