Cetuximab + pertuzumab is a combination of two monoclonal antibodies targeting different aspects of the HER signaling pathway. Cetuximab binds to EGFR (HER1), preventing ligand binding and receptor activation, while pertuzumab binds to the dimerization domain (subdomain II) of HER2, blocking its heterodimerization with other HER family receptors. The combination was investigated for treatment of cetuximab-resistant KRAS wild-type metastatic colorectal cancer but was found to have intolerable toxicity despite showing some antitumor activity.
