Cetuximab + platinum + gemcitabine is a combination regimen used primarily in the treatment of advanced or metastatic cancers, including non-small cell lung cancer (NSCLC) and head and neck squamous cell carcinoma (HNSCC). Cetuximab is a monoclonal antibody that targets the extracellular domain of the epidermal growth factor receptor (EGFR), blocking ligand binding and downstream signaling pathways involved in tumor proliferation, survival, angiogenesis, and metastasis. Platinum agents (such as cisplatin or carboplatin) are DNA-damaging chemotherapeutics that form DNA crosslinks leading to apoptosis. Gemcitabine is a nucleoside analog that inhibits DNA synthesis by incorporation into DNA during replication, causing S-phase arrest and cytotoxicity. The combination aims to enhance antitumor efficacy through complementary mechanisms—cetuximab-induced G1 arrest/EGFR inhibition with chemotherapy-induced cytotoxicity[1][3][4][6]. This regimen has been studied in phase II/III trials for NSCLC and HNSCC.
