Cetuximab + savolitinib is an investigational combination therapy comprising two targeted anticancer agents. Cetuximab is a chimeric monoclonal antibody that inhibits the epidermal growth factor receptor (EGFR), blocking downstream signaling pathways involved in cell proliferation and survival. Savolitinib is a highly selective small molecule inhibitor of the mesenchymal epithelial transition factor (MET) tyrosine kinase, which plays a key role in tumor growth, metastasis, and resistance to EGFR inhibitors. This combination aims to overcome acquired resistance mechanisms in cancers—particularly non-small cell lung cancer (NSCLC)—where MET amplification or overexpression drives resistance to EGFR-targeted therapies. The rationale for combining these agents is based on preclinical and clinical evidence that dual inhibition of EGFR and MET can provide synergistic antitumor effects in tumors with co-activation of these pathways.
