Drug intelligence / Profile preview
The combination of cetuximab beta, irinotecan, fluorouracil, and leucovorin (often referred to as cetuximab beta plus FOLFIRI) is an antineoplastic regimen used for the treatment of colorectal cancer. Cetuximab beta (Enlituo, formerly CMAB009) is a recombinant chimeric monoclonal antibody targeting the epidermal growth factor receptor (EGFR), developed using a proprietary expression process to minimize immunogenic glycosylation and reduce hypersensitivity reactions. Irinotecan is a small molecule topoisomerase I inhibitor, fluorouracil is a thymidylate synthase inhibitor, and leucovorin acts as a biochemical sensitizer that stabilizes the thymidylate synthase-fluorodeoxyuridine monophosphate complex. Together, this combination provides synergistic antitumor activity by simultaneously blocking EGFR-mediated signaling pathways and disrupting DNA replication and repair mechanisms in rapidly dividing cancer cells.
