Overview

Cevostamab + iberdomide is an investigational combination regimen being studied primarily for use in relapsed or refractory multiple myeloma. **Cevostamab** is a bispecific monoclonal antibody that targets CD3 on T cells and Fc receptor-homolog 5 (FcRH5) on myeloma cells, bringing immune cells in proximity to myeloma cells to facilitate their destruction through immunologic cytotoxicity. **Iberdomide** is a small molecule classified as a cereblon E3 ligase modulator (CELMoD), structurally related to immunomodulatory drugs (IMiDs) and functions by recruiting the substrate receptors Ikaros (IKZF1) and Aiolos (IKZF3) for ubiquitin-mediated degradation. This results in anti-neoplastic, immunomodulatory, and pro-apoptotic effects. The combination is under evaluation for safety, tolerability, pharmacokinetics, and efficacy in patients with heavily pretreated multiple myeloma, especially those who have failed standard therapies[1][2][3][4].
