Overview

This is an **investigational combination therapy** comprised of: - **CF33-CD19**, an oncolytic chimeric poxvirus (engineered orthopoxvirus, also called onCARlytics) that selectively infects and replicates within tumor cells, while encoding a truncated, non-signaling human CD19 transgene. Infected tumor cells express CD19 on their surfaces, rendering them susceptible to further immune targeting. - **Blinatumomab**, a bispecific T cell engager (BiTE) monoclonal antibody that simultaneously binds CD3 (on T cells) and CD19 (on B cells or transduced tumor cells), facilitating immune-mediated cell lysis. - **Hydroxyurea**, a ribonucleotide reductase inhibitor and small molecule chemotherapeutic that inhibits DNA synthesis and is used for cytoreduction. The combination aims to "tag" solid tumor cells for recognition by CD19-targeted therapies by inducing CD19 expression (via CF33-CD19) and subsequently targeting those cells with blinatumomab-mediated T cell engagement. Hydroxyurea may be used as a chemotherapeutic adjunct for cytoreduction or to modulate the tumor/immune microenvironment. The regimen is under phase I clinical investigation for adults with advanced or metastatic solid tumors[1][3][5][7].
