**CF33-hNIS + modified FOLFOX** is an investigational combination regimen involving an *oncolytic virus*, CF33-hNIS (also known as VAXINIA), and a chemically modified version of the standard chemotherapy regimen FOLFOX. CF33-hNIS is a genetically engineered chimeric poxvirus designed to selectively infect, replicate within, and lyse cancer cells, simultaneously expressing the human sodium-iodide symporter (hNIS) to enable imaging, tracking, and potential radiotherapeutic add-ons[3][1]. The mechanism of CF33-hNIS involves direct oncolysis as well as immune-mediated antitumor activity via enhanced antigen presentation and recruitment of immune effector cells[1][3]. Modified FOLFOX is a variation of the classic FOLFOX regimen, containing oxaliplatin, 5-fluorouracil (5-FU), and leucovorin, which acts as a cytotoxic chemotherapy backbone and can enhance immunogenic cell death, synergizing with virotherapy. This combination is being evaluated mainly in clinical trials for the treatment of advanced solid tumors, such as metastatic gastrointestinal cancers. CF33-hNIS was co-developed by City of Hope and Imugene[1][3].
