This program involves the development of small molecule inhibitors targeting cyclic GMP-AMP synthase (cGAS), an enzyme that acts as a cytosolic DNA sensor. Upon binding to double-stranded DNA, cGAS produces the second messenger cyclic GMP-AMP (cGAMP), which activates the Stimulator of Interferon Genes (STING) pathway, leading to the production of Type I interferons and other pro-inflammatory cytokines. Dysregulation of this pathway is implicated in various autoimmune and inflammatory diseases, such as Aicardi-Goutières syndrome (AGS), systemic lupus erythematosus (SLE), and other interferonopathies. The Leuven Centre for Drug Design and Discovery (CD3) is developing these inhibitors to modulate the cGAS-STING axis for the treatment of inflammatory conditions. The project is currently in the preclinical stage.
