This investigational HIV-1 vaccine, developed by the University of Oxford's Jenner Institute in collaboration with the International AIDS Vaccine Initiative (IAVI), utilizes a heterologous prime-boost regimen to elicit broad T-cell immunity. The regimen, designated C1-M3M4, consists of a prime dose using the simian adenovirus vector **ChAdOx1.tHIVconsv1**, followed by a simultaneous boost with two modified vaccinia virus Ankara (MVA) vectors, **MVA.tHIVconsv3** and **MVA.tHIVconsv4**. These vectors deliver the **HIVconsvX** immunogens, which are bivalent mosaic proteins designed from six highly conserved regions of the HIV-1 Gag and Pol proteins. By targeting these conserved regions across multiple HIV-1 clades, the vaccine aims to provide broad protection and overcome the high genetic diversity of the virus. It has been evaluated in Phase 1 clinical trials (such as HIV-CORE 006) to assess safety and immunogenicity in healthy HIV-negative adults.
