Overview

Chemotherapy + avelumab + talazoparib is an investigational combination regimen that was evaluated for the first-line treatment of advanced ovarian cancer. The regimen consists of avelumab (an anti-PD-L1 monoclonal antibody), talazoparib (a PARP1/2 inhibitor), and platinum-based chemotherapy. The therapeutic strategy involved an initial phase of chemotherapy combined with avelumab to induce tumor cell death and increase antigen presentation, followed by a maintenance phase of avelumab and talazoparib to provide sustained immune activation and inhibit DNA damage repair. The Phase 3 JAVELIN Ovarian PARP 100 trial (NCT03642132) investigating this combination was terminated in 2019 by developers Pfizer and Merck KGaA after interim results from a related study suggested the combination would not meet its primary efficacy endpoints.
