Chemotherapy + GI-4000 refers to the combination of standard cytotoxic chemotherapy regimens (most commonly gemcitabine) with GI-4000, a yeast-based therapeutic cancer vaccine. GI-4000 is composed of four different Tarmogens (GI-4014, GI-4015, GI-4016, and GI-4020), each expressing a different mutant Ras protein antigen corresponding to the most common RAS mutations found in solid tumors such as pancreatic and colorectal cancers. The mechanism of action for GI-4000 involves stimulating an immune response—specifically activating T cells—to recognize and attack tumor cells harboring these mutant Ras proteins. This combination has been investigated primarily as adjuvant therapy in patients with resected pancreatic cancer and also in metastatic colorectal cancer settings. The goal is to enhance anti-tumor immunity following reduction of tumor burden by surgery or chemotherapy[1][2][3][4][5].
