Talazoparib in combination with chemotherapy is an investigational treatment approach that pairs a PARP (poly ADP-ribose polymerase) inhibitor with various chemotherapy agents. Talazoparib works through dual mechanisms: inhibiting the PARP enzyme and trapping PARP at DNA damage sites, which prevents DNA repair and leads to cancer cell death. When combined with chemotherapy agents that cause DNA damage (such as temozolomide, topotecan, or gemcitabine), the combination may produce synergistic anti-tumor effects. This approach is being studied in various cancer types, particularly in pediatric patients with relapsed or refractory cancers including acute myeloid leukemia (AML) and solid tumors.
