Overview

Chidamide + azacitidine is a combination epigenetic therapy that pairs a histone deacetylase inhibitor (chidamide) with a DNA methyltransferase inhibitor (azacitidine). This dual epigenetic regulating treatment shows synergistic effects in treating hematological malignancies. Chidamide specifically targets HDAC1, 2, 3, and 10, preventing histone deacetylation and promoting gene expression. Azacitidine works through cytotoxicity (incorporating into RNA/DNA) and hypomethylation (inhibiting DNA methyltransferase). The combination has shown particular efficacy in peripheral T-cell lymphomas, especially angioimmunoblastic T-cell lymphoma (AITL), and is being investigated for acute myeloid leukemia and other hematological cancers.
