This is a combination regimen of three drugs: - **Chidamide (also known as tucidinostat):** A selective benzamide class histone deacetylase inhibitor (HDACi) that primarily targets subtypes 1, 2, and 3 of class I HDAC and subtype 10 of class IIb HDAC. It acts as an epigenetic modulator to alter gene expression in tumor cells[1][5]. - **Tucidinostat:** An alternative name for chidamide; both refer to the same active pharmaceutical ingredient[3][5]. - **Bevacizumab:** A recombinant humanized monoclonal antibody targeting vascular endothelial growth factor (VEGF), thereby inhibiting angiogenesis and tumor blood vessel formation[7][10]. The combination is being investigated for the treatment of advanced or metastatic microsatellite stable/proficient mismatch repair (MSS/pMMR) colorectal cancer that has failed at least two lines of standard therapy. The rationale is to combine epigenetic modulation with antiangiogenic therapy to enhance immune response within the tumor microenvironment. Clinical trials have shown improved progression-free survival and response rates when this triplet regimen is used compared to doublet regimens lacking bevacizumab[2][3].
