Overview

Chidamide + Zimberelimab is an investigational combination therapy consisting of two agents with complementary mechanisms of action for the treatment of cancer. Chidamide is a selective histone deacetylase inhibitor (HDACi) that modulates epigenetic regulation, leading to increased acetylation of chromatin histones, altered gene expression, cell cycle inhibition, and apoptosis in tumor cells. It also enhances antitumor immunity by upregulating tumor antigen presentation and immune co-stimulatory molecules while inhibiting immunosuppressive cells. Zimberelimab is a humanized monoclonal antibody targeting programmed cell death protein 1 (PD-1), functioning as an immune checkpoint inhibitor to restore T-cell mediated antitumor responses. The combination aims to synergistically improve the efficacy of immunotherapy by both increasing tumor immunogenicity and blocking PD-1-mediated immune evasion. This regimen is under clinical investigation for metastatic triple-negative breast cancer and other solid tumors[1][2][3][4].
