The chimpanzee adenovirus + self-amplifying messenger RNA (samRNA) vaccine is a heterologous prime-boost immunotherapy platform developed by Gritstone bio. The platform utilizes a chimpanzee adenovirus serotype 68 (ChAd68) vector to prime the immune system, followed by a boost with samRNA. This combination is designed to induce high-titer neutralizing antibodies and a robust, durable CD8+ T-cell response against targeted antigens. In oncology, the platform is applied through the GRANITE (individualized neoantigens) and SLATE (shared neoantigens) programs to treat advanced solid tumors, often in combination with immune checkpoint inhibitors like nivolumab and ipilimumab. The technology has also been adapted for infectious diseases, notably in the CORAL program for COVID-19, where it delivers SARS-CoV-2 spike and non-spike antigens to provide broad and lasting protection.
