Chlordiazepoxide + amitriptyline is a fixed-dose combination medication containing two active ingredients: chlordiazepoxide, a benzodiazepine, and amitriptyline, a tricyclic antidepressant (TCA). This combination is primarily used to treat moderate to severe depression associated with anxiety. Chlordiazepoxide acts by enhancing the effect of gamma aminobutyric acid (GABA) at GABA-A receptors in the central nervous system, producing anxiolytic and sedative effects. Amitriptyline increases synaptic concentrations of norepinephrine and serotonin by inhibiting their reuptake at nerve terminals, thereby improving mood and alleviating depressive symptoms. The combination targets both mood regulation and anxiety symptoms[1][2][3][6].
