Overview

Chloroquine + docetaxel is a combination therapy consisting of the antimalarial and autophagy inhibitor chloroquine and the taxane-class chemotherapeutic agent docetaxel. This combination is being investigated primarily for its potential to overcome multidrug resistance (MDR) in cancer cells, particularly in advanced or metastatic breast cancer. Chloroquine inhibits autophagy—a survival mechanism that can allow cancer cells to resist chemotherapy—thereby sensitizing tumor cells to the cytotoxic effects of docetaxel. Docetaxel stabilizes microtubules, preventing cell division and promoting apoptosis in rapidly dividing tumor cells. Preclinical studies have shown that co-delivery of these agents enhances anticancer efficacy compared to either drug alone, especially against resistant cancer cell lines[1][4]. Clinical trials have demonstrated promising objective response rates with manageable toxicity profiles when this combination is used after failure of anthracycline-based chemotherapy[5].
