Overview

Chlorproguanil + dapsone + artesunate (CDA) was an experimental fixed-dose artemisinin-based combination therapy (ACT) developed for the treatment of acute uncomplicated *Plasmodium falciparum* malaria. Developed through a public-private partnership involving GSK, the Medicines for Malaria Venture (MMV), and the WHO Special Programme for Research and Training in Tropical Medicine (WHO TDR), CDA was designed to be an affordable and effective treatment for use in Africa. The combination utilized three small molecule drugs with distinct mechanisms: chlorproguanil (a dihydrofolate reductase inhibitor), dapsone (a sulfone that inhibits dihydropteroate synthase), and artesunate (an artemisinin derivative that induces rapid parasite clearance). Despite reaching Phase III clinical trials, the development program was discontinued in 2008 after data revealed that the drug caused unacceptable hematological toxicity, specifically post-treatment hemolytic anemia, in patients with glucose-6-phosphate dehydrogenase (G6PD) deficiency.
