Overview
Ciforadenant + atezolizumab is a combination immunotherapy regimen under investigation for the treatment of advanced solid tumors, including metastatic castration-resistant prostate cancer (mCRPC) and non-small cell lung cancer (NSCLC). Ciforadenant (also known as CPI-444) is an oral small molecule antagonist of the adenosine A2A receptor, which blocks adenosine-mediated immunosuppression in the tumor microenvironment. By inhibiting this pathway, ciforadenant aims to restore anti-tumor activity of T cells and other immune cells. Atezolizumab is a monoclonal antibody that targets programmed death-ligand 1 (PD-L1), preventing it from binding to PD-1 on T cells and thereby enhancing immune-mediated tumor cell killing. The combination seeks to synergistically enhance anti-tumor immunity by simultaneously blocking two distinct immunosuppressive pathways within tumors[4][5][6][8].
