Cytokine-induced killer (CIK) cells are a heterogeneous population of immune effector cells, primarily consisting of CD3+CD56+ T cells, generated by the ex vivo expansion of peripheral blood mononuclear cells in the presence of interferon-gamma, anti-CD3 antibodies, and interleukin-2. Developed by Beijing GoBroad Hospital, these cells are being evaluated in a Phase 2 clinical trial to treat children and young adults with relapsed or refractory acute B-lymphoblastic leukemia (r/r B-ALL) who exhibit early functional exhaustion of Chimeric Antigen Receptor T-cell (CAR-T) therapy. CIK cells combine the potent cytotoxicity of T cells with the non-major histocompatibility complex (MHC)-restricted tumor recognition capabilities of natural killer (NK) cells. The hospital's research program includes both standard CIK cells and mRNA-modified CIK cells, aiming to sustain anti-leukemic activity and improve event-free survival in patients where CAR-T cells have failed to persist or function effectively.
