Cilostazol + galantamine is a combination of two small molecule drugs investigated primarily for their potential synergistic effects in the treatment of Alzheimer's disease and related cognitive disorders. Cilostazol is a phosphodiesterase 3 (PDE3) inhibitor with antiplatelet and vasodilatory properties, commonly used to treat intermittent claudication. Galantamine is an acetylcholinesterase inhibitor approved for mild to moderate dementia associated with Alzheimer's disease; it also acts as an allosteric modulator of nicotinic acetylcholine receptors. Clinical studies suggest that the combination may provide additional benefits over monotherapy in improving cognition and activities of daily living in patients with Alzheimer's disease, particularly those with comorbid cerebrovascular conditions or asymptomatic lacunar infarction[1][4][6]. The mechanism involves both enhancement of cholinergic neurotransmission (galantamine) and improved cerebral blood flow/neuroprotection (cilostazol). This combination remains investigational for neurodegenerative indications.
