Drug intelligence / Profile preview
Ciltacabtagene autoleucel + teclistamab is a combination regimen pairing ciltacabtagene autoleucel (cilta-cel, brand name CARVYKTI), a BCMA-directed CAR-T cell therapy, with teclistamab (brand name TECVAYLI), a bispecific T-cell engager targeting BCMA and CD3. Cilta-cel involves genetically modifying patient-derived autologous T cells to express a CAR with two distinct BCMA-binding domains for high avidity, promoting T-cell activation, expansion, and elimination of BCMA-expressing myeloma cells following a single intravenous infusion after lymphodepletion. Teclistamab recruits T cells to BCMA-positive plasma cells via subcutaneous administration. Developed primarily by Johnson & Johnson Innovative Medicine (Legend Biotech for cilta-cel manufacturing), this pairing is under preclinical and early clinical investigation to optimize sequencing or combination strategies for relapsed/refractory multiple myeloma (RRMM), leveraging complementary T-cell engaging mechanisms to enhance durable responses and address antigen loss or T-cell exhaustion.[1][2][3][9]
