Overview

Cinryze + rHuPH20 is a combination of two biologic agents developed for the prevention of hereditary angioedema (HAE) attacks. Cinryze is a plasma-derived C1 esterase inhibitor (human), approved for intravenous use to prevent angioedema attacks in patients with HAE. rHuPH20, or recombinant human hyaluronidase, is an enzyme that temporarily degrades hyaluronan in the subcutaneous tissue, facilitating the absorption and dispersion of co-administered drugs. The combination was investigated to enable subcutaneous administration of Cinryze by improving its bioavailability and patient convenience compared to intravenous infusion. Clinical trials showed that while the combination could be administered subcutaneously and reduced attack rates at higher doses, development was discontinued due to the emergence of non-neutralizing anti-rHuPH20 antibodies in some patients; these antibodies were not associated with adverse clinical effects but led to early termination as a precaution[4][5][7][8].
