Cisplatin works by forming intra- and inter-strand DNA cross-links, which inhibit DNA replication and transcription, leading to apoptosis. 5-fluorouracil acts as an antimetabolite that irreversibly inhibits thymidylate synthase, thereby disrupting the synthesis of thymidine monophosphate and subsequently DNA synthesis.
