cisplatin + 5-fluorouracil + tegafur-uracil

A combination chemotherapy regimen developed and evaluated by the Japan Clinical Oncology Group (JCOG) for the adjuvant treatment of serosa-positive gastric cancer. The protocol, specifically studied in the JCOG9206-2 phase III trial, consists of a complex administration schedule: intraperitoneal cisplatin administered immediately after surgery, followed by intravenous cisplatin and 5-fluorouracil (5-FU) on postoperative days 14–16, and a long-term maintenance phase of daily oral UFT (a combination of tegafur and uracil) for 12 months. Cisplatin functions as a platinum-based agent that creates DNA cross-links to inhibit replication, while 5-FU and the tegafur component of UFT act as antimetabolites by inhibiting thymidylate synthase. Despite the intensive approach, the JCOG9206-2 trial demonstrated that this regimen did not significantly improve overall survival or relapse-free survival compared to surgery alone in patients with macroscopically serosa-positive gastric cancer.
