This is a combination regimen consisting of three established anticancer agents—cisplatin, capecitabine, and trastuzumab—plus the investigational agent MM‑111. Cisplatin is a platinum-based chemotherapy that forms DNA crosslinks and induces apoptosis in rapidly dividing cells. Capecitabine is an oral prodrug of 5-fluorouracil, inhibiting thymidylate synthase and interfering with DNA synthesis. Trastuzumab is a monoclonal antibody targeting the human epidermal growth factor receptor 2 (HER2/ERBB2), blocking downstream signaling and mediating antibody-dependent cellular cytotoxicity in HER2-positive cancers[1][4][8]. MM‑111 is a bispecific recombinant fusion protein designed to inhibit both HER2 and HER3 receptors by binding to their extracellular domains, thereby disrupting neuregulin-mediated signaling pathways critical for tumor cell survival in certain cancers[6]. This combination has been investigated primarily for advanced or metastatic HER2-positive gastric or gastroesophageal junction adenocarcinoma.
