This is a combination regimen consisting of four drugs used in oncology, particularly for the treatment of advanced or metastatic non-small cell lung cancer (NSCLC) and potentially other solid tumors. The regimen includes: - **Cisplatin** and **carboplatin**, both platinum-based chemotherapeutic agents that cause DNA crosslinking and inhibit DNA synthesis, leading to apoptosis in rapidly dividing cells. - **Pemetrexed**, an antifolate antineoplastic agent that inhibits multiple folate-dependent enzymes involved in the synthesis of thymidine and purine nucleotides, thereby disrupting DNA replication and cell division. - **Pembrolizumab**, a monoclonal antibody targeting programmed death receptor 1 (PD-1), which blocks its interaction with PD-L1/PD-L2, enhancing T-cell mediated immune responses against tumor cells. This combination leverages cytotoxic chemotherapy to reduce tumor burden while pembrolizumab stimulates the immune system to recognize and attack cancer cells. It is primarily indicated for first-line treatment of metastatic nonsquamous NSCLC without EGFR or ALK genomic aberrations[4][6]. Pembrolizumab is developed by Merck (MSD outside North America).
