Overview

This multi-modal neoadjuvant chemo-immunotherapy regimen is being developed by Roswell Park Comprehensive Cancer Institute for the treatment of advanced-stage (III-IV) ovarian cancer. The regimen combines standard-of-care chemotherapy, including intraperitoneal cisplatin and intravenous paclitaxel, with a chemokine modulatory (CKM) regimen and an autologous alpha-DC1 dendritic cell vaccine. The CKM component consists of rintatolimod (a TLR3 agonist) and interferon-alpha, which are administered to reprogram the tumor microenvironment by upregulating chemokines such as CCL5, CXCL9, and CXCL10. This reprogramming is intended to enhance the infiltration of tumor-killing cytotoxic T lymphocytes induced by the autologous dendritic cell vaccine. Oral celecoxib is included to inhibit COX-2-mediated immunosuppression. The regimen is currently being evaluated in Phase 1/2 clinical trials to determine its safety and efficacy in achieving complete pathologic responses.
