Cisplatin + cyclophosphamide is a combination chemotherapy regimen being investigated for the treatment of metastatic triple-negative breast cancer (TNBC). In this specific protocol, developed by the Centre Jean Perrin, cyclophosphamide is administered using a metronomic dosing schedule—continuous, low-dose oral administration—rather than traditional high-dose bolus injections. This approach is combined with intravenous cisplatin. Cyclophosphamide acts as a DNA alkylating agent, while cisplatin is a platinum-based agent that creates inter- and intra-strand DNA cross-links. Together, they disrupt DNA replication and transcription, leading to cell cycle arrest and apoptosis. The metronomic use of cyclophosphamide is particularly aimed at providing sustained anti-angiogenic effects and overcoming resistance in patients previously treated with anthracyclines and taxanes.
