Cisplatin + docetaxel + doxorubicin is a combination chemotherapy regimen composed of three cytotoxic agents frequently used in the treatment of various solid tumors, including breast cancer. Each component has a distinct mechanism of action: - Cisplatin is a platinum-based compound that forms DNA crosslinks, leading to apoptosis by inhibiting DNA synthesis and function. - Docetaxel is a taxane that stabilizes microtubules and prevents their depolymerization, thereby inhibiting cell division. - Doxorubicin is an anthracycline antibiotic that intercalates into DNA, inhibits topoisomerase II, and generates free radicals causing DNA damage. This combination exploits different mechanisms to maximize antitumor efficacy. Sequential or concurrent administration has been studied in locally advanced breast cancer and other malignancies[1]. The regimen can be associated with significant toxicities such as neutropenia, mucositis, myalgia, bone pain, vomiting, renal impairment (from cisplatin), cardiotoxicity (from doxorubicin), and hypersensitivity reactions (notably from docetaxel)[1][4].
