This is a combination regimen consisting of three drugs: - Cisplatin, a platinum-based chemotherapeutic agent that forms DNA crosslinks, leading to apoptosis in rapidly dividing cells. - Docetaxel, a taxane-class chemotherapeutic that stabilizes microtubules and inhibits cell division. - Esomeprazole, a proton pump inhibitor (PPI) primarily used to reduce gastric acid secretion but also shown to sensitize tumor cells to chemotherapy by inhibiting vacuolar (H+)-ATPases and modulating the tumor microenvironment. The combination has been studied for its potential synergistic effects in cancer therapy. Esomeprazole may enhance the cytotoxicity of cisplatin and docetaxel by increasing tumor cell sensitivity through inhibition of pH-regulating mechanisms (such as vacuolar H+-ATPase), suppression of the AKT/mTOR signaling pathway, and promotion of apoptosis. Clinical studies have explored this triple regimen particularly in metastatic breast cancer and preclinical models suggest benefit in ovarian cancer and non-small-cell lung cancer[1][2][3][4].
