This is a combination chemotherapy regimen consisting of three cytotoxic agents—cisplatin, docetaxel, and etoposide. Each drug has a distinct mechanism of action: - Cisplatin is a platinum-based compound that forms DNA crosslinks, leading to apoptosis in rapidly dividing cells. - Docetaxel is a taxane that stabilizes microtubules and inhibits their depolymerization, thereby disrupting mitosis. - Etoposide is a topoisomerase II inhibitor that prevents DNA unwinding and replication. This combination has been investigated primarily for the treatment of advanced or metastatic non-small cell lung cancer (NSCLC), as well as other solid tumors. The rationale for combining these agents lies in their complementary mechanisms and non-overlapping toxicity profiles. Clinical trials have evaluated various doublet combinations (e.g., cisplatin + docetaxel or cisplatin + etoposide) with demonstrated efficacy in NSCLC[1][2][5]. However, the triple combination may be used in select protocols or investigational settings where maximal cytotoxic effect is desired.
