This is a combination regimen consisting of three drugs: - **Cisplatin** (a platinum-based chemotherapy agent that causes DNA crosslinking and apoptosis in rapidly dividing cells) - **Durvalumab** (a monoclonal antibody targeting PD-L1, acting as an immune checkpoint inhibitor to enhance anti-tumor immune response) - **Pemetrexed** (an antifolate antineoplastic agent that inhibits folate-dependent enzymes involved in the synthesis of DNA and RNA) The combination is used primarily for the treatment of unresectable malignant pleural mesothelioma and has also been investigated in non-small cell lung cancer. In clinical trials, this regimen demonstrated improved overall survival compared to historical controls using cisplatin and pemetrexed alone. The addition of durvalumab leverages immunotherapy alongside standard chemotherapy, aiming to improve outcomes by both direct cytotoxicity and immune-mediated tumor cell killing[1][2][3].
