This is a combination chemotherapy regimen consisting of three cytotoxic agents: cisplatin, etoposide, and fluorouracil (also known as 5-fluorouracil or 5-FU). Each drug has a distinct mechanism of action targeting cancer cell proliferation. Cisplatin is a platinum-based compound that forms DNA crosslinks, inhibiting DNA synthesis and function. Etoposide inhibits topoisomerase II, leading to DNA strand breaks and apoptosis. Fluorouracil is an antimetabolite that interferes with thymidylate synthase, disrupting DNA synthesis. The combination has been studied primarily in advanced non-small cell lung cancer (NSCLC), often referred to as the FED regimen[1][2][4]. It has also been explored in other solid tumors such as gastric cancer when combined with additional agents[3]. The main toxicities are hematologic and gastrointestinal.
