This is a combination regimen consisting of three agents: - **Cisplatin** is a platinum-based chemotherapeutic that forms DNA crosslinks and inhibits DNA synthesis and function. - **Etoposide** is a topoisomerase II inhibitor that causes DNA strand breaks by preventing the religation of double-stranded breaks induced by the enzyme. - **Interferon alpha** (including recombinant forms such as IFN-alpha2a or IFN-alpha2b) is an immunomodulatory cytokine with direct antiproliferative effects on tumor cells and immune-stimulating properties. It enhances antitumor immunity through activation of immune effector cells and modulation of antigen presentation. This combination has been studied primarily in small cell lung cancer (SCLC), especially extensive disease SCLC, where it was used as induction therapy followed by maintenance interferon[1]. Preclinical studies suggest that interferon alpha can potentiate the cytotoxic effects of etoposide more than cisplatin in some cancer cell lines[2]. The regimen aims to combine direct cytotoxicity from chemotherapy with immunomodulation from interferon to improve outcomes in aggressive cancers.
