This is a combination regimen consisting of three agents: - **Cisplatin** is a platinum-based chemotherapeutic classified as an alkylating agent. It exerts its antineoplastic effects by cross-linking DNA, thereby inhibiting DNA replication and transcription, leading to cell death. Cisplatin is cell cycle-nonspecific and disrupts DNA function through multiple mechanisms including direct cross-linking of guanine bases[5]. - **Etoposide** is an epipodophyllotoxin derivative that inhibits the enzyme DNA topoisomerase II, resulting in double-strand breaks in DNA and inhibition of cell division. Etoposide acts primarily during the S and G2 phases of the cell cycle[7]. - **Interferon gamma (IFN-γ)** is a type II interferon cytokine with immunomodulatory, antiproliferative, pro-apoptotic, antiangiogenic, and antitumor properties. It enhances antigen presentation on tumor cells via upregulation of MHC molecules and stimulates immune effector functions such as cytotoxic T lymphocyte activity[6][8][9]. This combination has been investigated in clinical trials for cancer therapy—most notably for non-small cell lung cancer—where IFN-gamma was used as an immunomodulatory adjunct to standard chemotherapy with cisplatin and etoposide[1][4]. The rationale was to enhance antitumor immunity alongside direct cytotoxic effects.
