This regimen is a multi-agent, multimodal cancer treatment protocol combining the chemotherapeutic agents cisplatin and etoposide with concurrent radiation therapy, followed by consolidation chemotherapy with docetaxel and supportive care using pegfilgrastim. Cisplatin is a platinum-based compound that forms DNA crosslinks, leading to apoptosis in rapidly dividing cells and also acts as a radiosensitizer to enhance the effects of radiation therapy. Etoposide inhibits topoisomerase II, causing DNA strand breaks and cell death. Radiation therapy targets tumor cells directly through ionizing radiation. After initial chemoradiation, docetaxel—a taxane that stabilizes microtubules and prevents cell division—is administered as consolidation therapy to further reduce residual disease. Pegfilgrastim is a long-acting granulocyte colony-stimulating factor (G-CSF) used prophylactically to reduce the risk of neutropenia associated with intensive chemotherapy[1][4][6]. This regimen has been studied primarily in patients with locally advanced or inoperable non-small cell lung cancer (NSCLC) but may be adapted for other solid tumors.
